Genetic family trees.
Distant relationships, such as cousins, great-uncles, or 2nd cousins, can be supported by DNA testing. Paternal and maternal line inheritance patterns to support or exclude an extended family relationship. This type of DNA testing is similar to that used for genetic genealogy and family group studies.
PATERNAL AND MATERNAL LINES
Y-Chromosomal DNA (Y-DNA)
Y-DNA appears in males only and always passes from father to son. Genetic genealogy primarily uses Y-DNA for surname studies, since Y-DNA corresponds to family names. Y-Chromosome DNA testing compares two Y-DNA profiles and finds a match (supports a close biological relationship) or no match (refutes a close biological relationship with a common paternal ancestor).
Participants in a Y-Chromosome comparison study must be related along a common paternal line as illustrated above. 2nd cousins or a great-uncle on the mother's side cannot participate in this test.
Mitochondrial DNA (mtDNA)
mtDNA appears in males and females and always passes from mother to child. Genetic genealogy primarily uses mtDNA for family group studies, where participants share common maternal ancestry. Mitochondrial DNA testing compares two mtDNA profiles and finds match (supports a close biological relationship) or no match (refutes a close biological relationship with a common maternal ancestor).
Participants in an mtDNA comparison study must be related along a common maternal line as illustrated above. 2nd cousins or a great-uncle on the father's side cannot participate in this test.
DISTANT RELATIONSHIPS
When comparing paternal or maternal lineage, DNA testing examines Y-chromosome (Y-DNA) or mitochondrial DNA (mtDNA) profiles from two (or more) individuals. If the DNA profiles are an exact match, the tested individuals share a common link within four generations and could be as closely related as siblings or cousins. However, they may share a more distant relationship such as 2nd cousins or great-grandparent to great-grandchild. If the profiles do not match, a close relationship, such as siblings, cousins, or aunt/uncle, is not possible.
Paternal line and maternal line comparison results are not as informative for close relationships as other types of DNA relationship testing. DNA tests for Y-DNA or mtDNA comparisons, however, may be the only DNA testing option in some cases.
